摘要
目的建立家兔血浆中乌头碱浓度的高效液相色谱(HPLC)测定方法。方法采用Symmetry shield RP18(5μm,4.6×250mm)色谱柱,以乙腈-0.1%甲酸(68∶32)为流动相,流速为0.8mL.min-1,检测波长为235nm。结果乌头碱在0.05~8μg.mL-1范围内成良好的线性关系(r2=0.999)。样品的回收率在95%~108%,日内和日间RSD均小于5%。结论本方法准确、灵敏、简便,适用于家兔血浆中乌头碱含量的测定分析。
OBJECTIVE To establish a HPLC method for determination of aconitine in rabbit plasma.METHODS The samples were analyzed on the column of Symmetry shield RP18(5μm,4.6×250mm).The mobile phase consisted of acetonnitrile-0.1% formic acid(68∶32) at the rate of 0.8mL·min-1.The detective wavelength was at 235nm.RESULTS Good linearity was obtained in the range of 0.05~8μg·mL-1(r2=0.999).The recovery rate was 95~108%,both the inter-day and intra day RSD were less than 5%.CONCLUSION The method was accurate,sensitive,simple,and suitable for the deternination of aconitine in rabbit plasma.
