高效液相色谱法测定复方虎杖紫草油中虎杖苷和白藜芦醇的含量

Determination of polydatin and resveratrol in Fufang Huzhang Zicao oil by HPLC

目的建立同时测定复方虎杖紫草油中虎杖苷和白藜芦醇含量的方法。方法采用Zorbax Eclipse XDB-C18(150×4.6,5μm);流动相:乙腈-0.1%冰醋酸溶液,检测波长:307nm。结果虎杖苷和白藜芦醇分别在0.026～0.520mg.mL-1(r=0.9998)和0.017～0330mg.mL-1(r=0.9998)浓度范围内有良好的线性关系,平均加样回收率分别为99.1%,98.3%;RSD%均小于2.0%。结论此方法操作简便、专属性强、灵敏度高、重现性好,可用于该制剂的质量控制。

OBJECTIVE To establish a method for determination of polydatin and resveratrol in Fufang Huzhang Zicao oil.METHODS The HPLC system consisting the Zorbax Eclipse XDB-C18（150×4.6,5μm） column,the mobile phase consisted of Acetonitrile-0.1% HAc,the UV detector was set at 307nm.RESULTS The liner response range was 0.026～0.520mg·mL-1（r=0.9998） and 0.017～0330mg·mL-1（r=0.9998）,respectively.The average recovery of polydatin and resveratrol were 99.1% and 98.3%,respectively,RSD2.0%.CONCLUSION The assay demonstrated that the method had adequate and selectively to measure the concentrations of accurate,repeatable,it can be applied in quantitative determination of polydation and resveratrol in Fufang Huzhang Zicao oil.
