摘要 为了实现大花蕙兰优良个体的快速繁殖,本文采用外源激素配合肌醇与椰汁的方法研究了大花蕙兰茎段的不定芽分化、增殖及试管苗的生根。结果表明:MS+6-BA 1.5 mg/L+NAA 0.5 mg/L+肌醇100 mg/L+活性炭1 g/L+蔗糖3﹪有利于大花蕙兰不定芽的分化,并可实现一步成苗;在1/2 MS+6-BA 0.3 mg/L+NAA 0.7 mg/L+椰汁100 mL/L+肌醇100 mg/L+活性炭1 g/L+蔗糖3﹪培养基中,培养60 d,生根率可达100﹪,试管苗生长健壮;培养基中加肌醇可以显著促进壮苗生根。但是,培养基中单独加入椰汁对试管苗不定芽与根的分化和生长有抑制作用。 Effects of exogenous hormone and additives on differentiation of buds, rooting of Cymbidium hybridium plantlets were studied. The results show that the medium MS+ 6-BA 1.5 mg/L + NAA 0.5 mg/L + cyclohexanhexol 100 mg/L + active carbon 1 g/L + sucrose 3 % was conducive to differentiation and multiplication, and rooting. The rootage rate of plantlets reached 100 % in the medium of 1/2 MS+6-BA 0.3 mg/L + NAA 0.7 mg/L + coconut juice 100 mL/L + cyclohexanhexol 100 mg/L + active carbon 1 g/L + sucrose 3 % after the explants were cultured for 60 days. Coconut juice inhibited differentiation of buds and rooting, though cyclohexanhexol advanced them obviously.
