PD-ECGF表达与肾癌的临床病理及预后因素的关系 被引量:2

摘要 目的:试图揭示血小板源性内皮细胞生长因子(PD-ECGF)在肾癌组织中的表达规律,探讨其与肿瘤增殖、肿瘤血管生成、临床病理及预后之间的关系。方法:用逆转录聚合酶链反应(RT-PCR)检测87例肾癌组织标本PD-ECGF mRNA的表达;用免疫组织化学法检测PCNA及CD34在肿瘤中的表达,计算微血管密度(MVD),并结合临床和病理资料进行分析。结果:肾癌中PD-ECGF、PCNA及CD34表达随病理级别的增高而表达明显增强(P<0.01),而与患者的性别、年龄、肿瘤部位、肿瘤直径无关(P>0.05);PD-ECGF的表达与PCNA的表达及MVD之间显著相关;Cox回归多因素分析显示病理级别、淋巴转移、PD-ECGF、PCNA和MVD 5个因素对肾癌患者生存期的影响具有统计学意义。结论:PD-ECGF在肾癌的形成与发展过程中起重要作用,它可促进肿瘤增殖及瘤组织内微血管的发生,并对其预后判断有指导意义。 Objective To investigate the expression of platelet-derived endothelial cell growth factor(PD-ECGF)mRNA in renal carcinoma and their relationship with tumor proliferation,angiogenesis,clinical pathology and prognosis.Method The expression of PD-ECGF mRNA in the tumor tissues of 87 patients with renal carcinoma were detected by RT-PCR;immunohistochemistry technique was used to test the expression of proliferating cell nuclear antigen(PCNA) protein and the microvessel density(MVD) was determined by immunohistochemistry with monoclonal antibody against CD34.Results Statistical significant difference of PD-ECGF,PCNA and CD34 expressions was observed in different pathologic grades of renal carcinoma and the various postoperative survival states of these patients.The expression of PD-ECGF,PCNA and CD34 was not related to sex,age,position of tumor and diameter of tumor.The expression of PD-ECGF was related to the expression of PCNA and the MVD in the tumor tissues of these patients.Pathological grade,lymph node metastases,PD-ECGF,PCNA,MVD had a significant influence on the survival time.Conclusion PD-ECGF plays an important role in the formation and progression of the renal carcinoma and can promote tumor proliferation and intratumoral microvascular formation,and is closely related to the prognosis of the patients.
