依达拉奉对帕金森病小鼠模型黑质酪氨酸羟化酶及Caspase-3的影响 被引量:1

Effect of edaravone on tyrosine hydroxylase and caspase-3 in the substantia nigra of the Parkinson's disease mouse model

摘要 目的探讨依达拉奉对帕金森病(PD)小鼠黑质多巴胺神经元的保护作用。方法 30只健康雄性C57BL/6小鼠,随机分为正常对照组、模型组和治疗组。后两组给予腹腔注射百草枯和代森锰建立PD模型。建模成功后,治疗组给予依达拉奉腹腔注射2周,正常对照组给予同剂量生理盐水。观察3组小鼠行为学的变化,并用免疫组织化学法检测黑质TH、Caspase-3的表达。结果与治疗组比较,模型组小鼠爬杆时间明显延长,黑质TH阳性神经元数量显著降低,Caspase-3的表达显著增高。结论依达拉奉治疗可使PD小鼠运动能力增强,黑质TH阳性神经元数量增加,并降低Caspase-3的表达,有明显的神经保护作用。 Objective To investigate the protective effect of edaravone on nigrostriatal dopaminergic neurons in Parkinson's disease(PD) mice induced by paraquat(PQ) and maneb(MN).Methods 30 male C57BL/6 mice were randomly divided into the model group,the edaravone-treated group and the normal control group.After the PD mouse model was established by intraperitoneal injection of PQ and MN,mice in the edaravone-treated group were intraperitoneal administrated edaravone for 2 weeks.Motor coordination was detected by using the behavioral method,and expressions of tyrosine hydroxylase(TH) and caspase-3 in SNc by immunohistochemistry.Results Administration of edaravone significantly attenuated the impairment of behavioral performance,increased the number of TH-positive neurons,and decreased expression of caspase-3 in SNc.Conclusion Edaravone can improve the motor coordination of PD mice,increase the number of TH-positive neurons,and decrease expression of caspase-3.It has beneficial influence on neurons in PD mice.
