哈氏仿对虾溶菌酶序列的克隆及分析

摘要 利用RT—PCR以及RACE的方法在哈氏仿对虾中分离得到1个溶菌酶序列,全长711bp,5端非翻译区长度为40bp,3端非翻译区为208bp,包括加A信号序列AATAAA与多聚A结构,开放阅读框(ORF)462bp,编码153个氨基酸,其中信号肽长度为18个氨基酸,成熟肽长度为135个氨基酸,序列中具有8个半胱氨酸碱基、2个催化反应位点及1个Ca2+结合位点,表明该序列属于C型溶菌酶。该序列的获得,有助于为进一步研究溶菌酶的活性。 The cDNA of lysozyme was cloned from Parapenaeopsis hardwickii by RT-PCR and RACE techniques. The complete cDNA sequence of lysozyme is 711 bp, consisting of a 5-UTR of 40 bp and 3- UTR of 209 bp with a polyadenylation signal sequence AATAAA and a poly(A) tail, the length of ORF is 462 bp encoded a polypeptides of 153 amino acid with a signal peptide of 18 amino acid and a mature protein 135 amino acids. This lysozyme presumed to be a chicken-type lysozyme for it possesses two catalytic sites, eight cysteine residues and one Ca2+ binding site. This work provides a basis for further studying of activity of the lysozyme.
