厌氧氨氧化细菌的分子生物学鉴定

摘要 采用分子生物学方法从厌氧氨氧化污泥中提取细菌总DNA,使用特异性引物进行PCR扩增。扩增产物经过克隆、测序后,得到厌氧氨氧化菌部分16S rDNA序列(长度为838 bp)。将得到的序列构建系统发育树进行分析。结果表明,厌氧氨氧化污泥中的厌氧氨氧化细菌与Anaerobic Ammonium-Oxidizing Planctomycete(AJ131819)细菌在进化上关系最近,并将该类细菌暂命名为Anaerobic Ammonium-Oxidizing Planctomycete ASBBR Gene。 The anaerobic ammonium oxidation(ANAMMOX) sludge was enriched in an anaerobic sequencing biofilm batch reactor(ASBBR).Total DNA of the bacteria was extracted from the anaerobic ammonium-oxidizing sludge by a molecular biology method,and was then amplified by the polymerase chain reaction(PCR)with a specific primer.After the PCR product was cloned and sequenced,the partial 16S rDNA sequence of ANAMMOX bacterium(length:838 bp)was obtained,and the sequence was analyzed by the phylogenetic tree.The results show that the cultivated ANAMMOX bacterium greatly closes to the anaerobic ammonium-oxidizing planctomycete(AJ131819),thus the bacterium is named temporarily as anaerobic ammonium-oxidizing planctomycete ASBBR gene.
