摘要
从酿酒酵母A_(364A)菌株制备原生质体,经超声波处理、磷酸纤维素P11吸附分离和DEAE-Sephadex A25层析等步骤,获得四个依赖于DNA的RNA聚合酶活性峰。根据层析行为和对α-鹅膏蕈碱的敏感性,确定四个活性峰依次分别为RNA聚合酶IA、IB、Ⅱ和Ⅲ。
The four active peaks of DNA-dependent RNA polyraerase were obtained by means of protoplast preparation of Saccharomyces cerevisiae A364A, sonication, batchwise adsorption on P11 phosphocelulose and chromatography on DEAE-Sephadex A 25. According to their chromatography behaviours and sensitivities to α-amanitin, the four peaks were identified with DNA-dependent RNA po-lymerase IA, IB, Ⅱ and Ⅲ in their given order.
