复方黑骨藤提取物对佐剂性关节炎大鼠滑膜组织中TNF-α、MMP-9和TIMP-1的影响 被引量:17

摘要 目的探讨复方黑骨藤50%乙醇提取物对类风湿性关节炎的治疗作用机理。方法构建大鼠完全弗式佐剂性(AA)关节炎模型,通过免疫组化法检测大鼠踝关节滑膜组织中肿瘤坏死因子-α(TNF-α),基质金属蛋白酶-9(MMP-9)和组织金属蛋白酶抑制剂-1(TIMP-1)的含量。结果空白对照组、阳性对照组及复方黑骨藤提取物实验组大鼠滑膜组织中TNF-α、MMP-9及TIMP-1的阳性表达均与模型对照组存在显著或极显著性差异。结论复方黑骨藤提取物能显著减轻佐剂性关节炎大鼠关节肿胀,抑制TNF-α及MMP-9的表达,增强TIMP-1的表达,调整MMP-9/TIMP-1的失衡,从而减轻关节炎的症状和软骨破坏,对实验性类风湿关节炎的滑膜增生具有显著的抑制作用。 Objective To investigate the therapeutic effect of Periploca forrestil Schltr.Compound extracts(PFSCE)extracted by 50% ethanol in the animal model of rheumatiod arthritics and mechanism. Methods To build the complete adjuvant arthritis(AA) model in rats,and then to detect the contents of tumor necrosis factor-α(TNF-α),matrix metalloproteinase-9(MMP-9)and tissue inhibitor-1 of metalloproteinas(TIMP-1)in synovial tissues of rats ankles by immunohistochemistry. Results By comparing the contents of TNF-α,MMP-9 and TIMP-1,there was difference or significant difference between blank control group(or positive control group,or the group of PFSCE) and model group. Conclusion The PFSCE can significantly reduce joint swelling in rats induced by adjuvant arthritis,inhibit the expression of TNF-α and MMP-9,enhance the expression of TIMP-1 and adjust the MMP-9/TIMP-1 imbalance,alleviate the symptoms of arthritis and cartilage destruction,thereby the PFSCE can significantly inhibit synovial proliferation of experimental rheumatoid arthritis.
