摘要
目的评价大豆苷元-壳聚糖长效微球对小鼠去卵巢骨质疏松的作用。方法建立小鼠去卵巢骨质疏松模型,测定给药后小鼠体质量和子宫重量系数,并取股骨进行石蜡切片HE染色观测。结果小鼠去卵巢后3周出现骨质疏松症状,所有实验组小鼠体质量无明显增重。与假手术组相比,去卵巢后子宫重量系数明显减小(P<0.01);肌肉注射高剂量大豆苷元-壳聚糖微球1次后,第3周和第5周子宫重量系数有较大幅度的增加(分别为P<0.05,P<0.01),明显高于每日灌胃大豆苷元混悬液组。与模型组相比,微球给药组可见骨小梁分布较密,骨皮质呈不明显蚕食样改变,骨髓腔较密实;而大豆苷元混悬液灌胃给药组无明显差别。结论与大豆苷元混悬液灌胃给药相比,大豆苷元-壳聚糖微球经肌肉注射有明显的改善骨质疏松症状作用,提示其在体内生物利用度得到提高。
Objective To evaluate the effect of daidzein-chitosan long sustained-microspheres on osteoporosis in ovariectomized mice. Methods The model of ovariectomized ICR mice was built successfully.Body weight and uterus index were measured,paraffin slides of the ovariectomized ICR mice femur were stained by HE. Results The osteoporosis symptoms happened after three weeks by ovariectomization.All groups had no change in body weight.There was significant decrease of uterus index after ovariectomization(P0.01) and increase of uterus index after three and five weeks(P0.01 and P0.05,respectively) by administering high dose of daidzein-chitosan microspheres only once,compared with sham operation group.The pathological section of femur by HE staining showed that high dose of daidzein-chitosan microspheres group increased the density and thickness of cortical bone. Conclusion Compared with daidzein i.g.administration,high dose of daidzein-chitosan microspheres improve the osteoporosis symptoms in ovariectomized mice,suggesting the increase of bioavailability in vivo.
