摘要 目的观察不同中医治法对LD模型大鼠多巴胺D2受体活性的影响。方法用6-羟基多巴胺注射于大鼠脑右侧黑质造成偏侧帕金森病(Parkinson,s disease,PD)模型,进一步腹腔注射左旋多巴(L-dihydroxy-phenylalanine,LD)制备LD模型。实验设立正常对照组、假手术组、PD模型对照组、LD模型组、平肝息风组、活血组、化痰组,连续治疗2,4,6周。采用放射性受体结合法测定大鼠纹状体多巴胺D2受体的最大结合容量(maximum binding capacity,Bmax)。结果正常对照组和假手术组大鼠纹状体多巴胺D2受体Bmax值在各时间段均无明显变化。LD模型组、PD模型对照组和各中药组的损毁侧在不同的时间段,多巴胺D2受体Bmax值均呈下降趋势,其中4周、6周与2周比较,差异均有显著性意义(P﹤0.01);6周与4周比较有下降趋势但差异不具有统计学意义(P﹥0.05)。LD模型大鼠健侧纹状体多巴胺D2受体Bmax值,亦有逐渐下降趋势,其中4周、6周与2周比较,差异均有统计学意义(P﹤0.05或P﹤0.01)。结论不同时间段各中药组可显著改善纹状体多巴胺D2受体活性,缓解左旋多巴的毒副反应。 Objective To observe the effect of different Chinese medicines on dopamine D2 receptor activity in PD rats. Methods The rat model of Parkinsons disease(PD) was established by injecting 6-hydroxydopami(6-OHDA) into the right substantia nigra of brain,then the model of LD was produced by injecting levodopa(LD) for 2 weeks.The rats were divided into normal control group,Sham group,PD model control group,LD model group,Calming liver wind group,Blood group and Phlegm group,continuous treatment for 2,4,6 weeks.The radioligand binding assay(RLBA) was used to measure the maximal binding capacity of receptor(Bmax). Results The Banax value of dopamine D2 receptors in rat stratum had no significant change between normal control group and sham operation group.The dopamine D2 receptor Bmax values in the lesioned side of LD model group,PD model control group and the Chinese groups were decreased in different time periods,and their differences were significant(P0.01)in 4 weeks and 6 weeks compared with 2 weeks;the difference was not statistically significant(P0.05) between 6 and 4 weeks.Uneffected Bmax value of striatal dopamine D2 receptors in LD rats was also gradually decreased,and their differences were significant(P0.05 or P0.01) in 4 weeks and 6 weeks as compared with 2 weeks. Conclusion The Chinese medicine groups can improve the activity of dopamine D2 receptor and alleviate side effects of levodopa at different time.
