摘要
目的:探讨人重组γ-干扰素(hrIFN-γ)对孕鼠血清血管内皮生长因子(VEGF)和耐热型碱性磷酸酶(HSAP)的影响。方法:采用妊娠第7~10天小鼠,不同浓度hrIFN-γ腹腔给药,2天后处死,用ELISA法检测小鼠血清中VEGF和HSAP的含量。结果:①干扰素与血管内皮生长因子:对照组数据明显低于实验第2、3、4组(P<0.05)。实验第2、3、4组之间无统计学差异(P>0.05),但随着干扰素浓度的升高,孕鼠血清中血管内皮生长因子呈上升趋势。②干扰素与耐热型碱性磷酸酶:实验第2、3、4组比对照组明显增高(P<0.05)。实验第2、3、4组之间无统计学差异(P>0.05),但随着干扰素浓度的升高,孕鼠血清中耐热型碱性磷酸酶水平呈上升趋势。③耐热型碱性磷酸酶与血管内皮生长因子:两组进行Pearson相关性分析显示,相关系数为0.534;两个指标间有相关性,一定剂量的IFN-γ能上调VEGF和HSAP的表达,且呈剂量依赖性。结论:hrIFN-γ对VEGF和HSAP的表达具有免疫调节作用。
Objective:To explore the effect of human recombinant γ-interferon on serum vascular endothelial growth factor(VEGF) and heat stable alkaline phosphatase(HSAP) levels of pregnant mice. Methods:The mice of 7-10 gestational days were selected and treated with transperitoneal injection of different concentrations of human recombinant γ-interferon,then they were killed after two days,enzyme-linked immunosorbent assay(ELISA) was used to detect the serum levels of VEGF and HSAP. Results:The serum level of VEGF in control group was significantly lower than those in the second group,the third group,and the fourth group(P0.05).There was no significant difference among the second group,the third group,and the fourth group(P0.05),the serum level of VEGF showed a increasing trend with the increase of serum human recombinant γ-interferon level.The serum levels of HSAP in the second group,the third group,and the fourth group were significantly higher than that in control group(P0.05).There was no significant difference among the second group,the third group,and the fourth group(P0.05),the serum level of HSAP showed a increasing trend with the increase of serum human recombinant γ-interferon level.The correlation coefficient between VEGF and HSAP was 0.534,there was a correlation.A certain dose of γ-interferon could upregulate the expressions of VEGF and HSAP,showing a dose-dependent manner. Conclusion:γ-interferon has an immunoregulative effect on the expressions of VEGF and HSAP.
