碳源对β-葡萄糖苷酶合成的影响研究 被引量:1

摘要 以黑曲霉(AspergiLLus niger NL-1)为产酶菌种,采用摇瓶发酵法,研究不同碳源对β-葡萄糖苷酶合成的影响。以水杨素为底物,以单位时间内催化水解水杨素产生葡萄糖的量表示酶活单位,测定酶活力。结果表明,麸皮为最佳的碳源,适宜的使用量为4%(质量分数)。以纯淀粉、微晶纤维素、葡萄糖、蔗糖、纤维二糖等为碳源酶活力很低,或不产酶。以麸皮为底物的最佳产酶时间为5 d,β-葡萄糖苷酶活力达到98.02 nkat/mL。纤维二糖对麸皮为碳源产β-葡萄糖苷酶无诱导作用。 Effects of carbon sources on the biosynthesis of β-Glucosidase by AspergiLLus niger NL-1. We investigated Salicin as the substrate was used go indicate activity in unit time. The results showed that bran was the best carbon source for β-Glucosidase production among the studied carbon sources, and it's preferable concentration was 4 %. Starch, microcrystalline cellulose, glucose, sucrose, cellobiose, etc. were not carbon sources for β-Glucosidase production. The best time for fermentation is 5 days, in the case, the highest activity of β-Glucosidase 98.02 nkat was obtained. CeUobiose as bran carbonsource hadn't been inducing effect.
