CO_2对宫颈癌细胞caveolin-1基因表达的影响及意义

摘要 目的探讨CO2人工气腹对宫颈癌Hela细胞caveolin-1(CAV-1)基因表达的影响及意义。方法宫颈癌Hela细胞用压力为8、16mm Hg的CO2作用4h,适时-荧光定量-PCR分别检测CO2未处理组及CO2处理后1、3、5d组caveolin-1基因mRNA的表达量。结果 CO2处理后宫颈癌细胞中caveolin-1mRNA明显降低,且caveolin-1基因的表达随CO2处理后时间的延长而降低。不同压力组间比较差异无统计学意义。结论 CO2人工气腹能抑制宫颈癌Hela细胞caveolin-1基因的表达,其可能是CO2气腹增强宫颈癌Hela细胞增殖能力的原因之一。 Objective To investigate the effect of CO2 on the expression of caveolin-1(CAV-1) gene of cervical cancer cells,and analyze the role of CO2 in the growth and metastasis of the cell.Methods The cervical cancer Hela cells were exposed to CO2 environment of 8 and 16 mm Hg pressure for 4 hours,then the cells were grouped with 1 day after treated、3 day after treated、5 day after treated and the untreated group.The real-time PCR was used to detect the expression of CAV-1 mRNA of hela cells in different groups.Results The expression of CAV-1 of Hela cells in the study groups was lower than that in the control group.In the study groups the expression of CAV-1 of Hela cells significantly reduced with the time extended after treated by CO2.Comparison of different pressure groups were not statistically significant.Conclusion CO2 environment can inhibit the expression of caveolin-1 gene of cervical cancer cells,which may be one of the reasons of enhanced proliferation ability of cervical cancer cells after CO2 pneumoperitoneum.
