响应面优化蒽酮-硫酸法测定水溶性沙棘叶多糖含量 被引量:10

Determination of Water Soluble Polysaccharides from Foliage in Hippophae Rhamnoides L.by Optimization of Anthrone-Sulfuric Acid Method with Response Surface Methodology

摘要 采用水提醇沉法从沙棘叶中提取水溶性沙棘叶多糖(WPFH),通过响应面优化蒽酮-硫酸显色,用分光光度法测定沙棘叶水溶性多糖的总糖含量。通过单因素和响应面法确定了最佳蒽酮-硫酸显色条件:显色温度为100℃,蒽酮试剂的用量为3.54mL,显色时间为10.95min。测定波长620nm,葡萄糖质量浓度在6.67—160μg/mL范围内与吸光度呈良好的线性关系,r=0.9993。该方法简单可行,平均回收率为100.36%,RSD为2.75%。精制WPFH换算因子f为1.266。 Water soluble polysaccharides from foliage in Hippophae Rhamnoides L.(WPFH) were extracted with hot water-ethanol method,and the total polysaccharides were determined by spectrophotometry at 620nm with anthrone-sulfuric acid method coloration,Single factor experiments and response surface analysis were applied to optimizing anthrone-sulfuric acid method.The optimum conditions of anthrone-sulfuric acid method were as follows:the anthrone-sulfuric acid dosage of 3. 54mL,coloration temperature of 100 C and coloration time of 10.95min.There was a good lineary between mass concentration and absorbence in the range of 6.67—160 pg/mL with r=0.9993.The method is simple and feasible.The average recovery was 100.36%with RSD of 2.75%.The conversion factor(f) was 1.266.
