重组人促红细胞生成素对大鼠肝脏缺血-再灌注损伤的保护作用 被引量:1

Protective effect of recombinant human erythropoietin on hepatic ischemia-reperfusion injury in rats

摘要 目的探讨重组人促红细胞生成素(rHuEPO)对大鼠肝脏缺血-再灌注(I-R)损伤的保护作用。方法将30只大鼠随机均分为假手术组(A组)、I-R模型组(B组)和rHuEPO 5000IU/kg处理组(C组)。建立70%大鼠肝脏I-R损伤模型,检测各组血清ALT、AST水平,HE染色观察组织学变化,Western blot及RT-PCR检测肝脏诱导型一氧化氮合酶(iNOS)和内皮型一氧化氮合酶(eNOS)的表达。结果与B组相比,C组iNOS mRNA和蛋白表达、ALT和AST水平均明显减少(P<0.05或P<0.01),肝细胞水肿、坏死减轻;而B组和C组间eNOS mRNA和蛋白表达差异无统计学意义(P>0.05)。结论 rHuEPO可能通过抑制iNOS表达,减轻大鼠肝脏I-R损伤。 Objective To explore the protective effect of recombinant human erythropoietin(rHuEPO) on the hepatic ischemia-reperfusion(I-R) injury in rats. Methods Thirty rats were equally randomized into three groups of A(sham operated),B(I-R model) and C(treated with rHuEPO 5000IU/kg before establishing I-R model). The 70% hepatic I-R injury model in rats was established,and the levels of ALT and AST were detected. The histological changes were observed by HE staining,and the expressions of induced nitricoxide synthase(iNOS) and endothelial nitric oxide synthase(eNOS) were examined by Western blot and RT-PCR. Results Compared with group B,the mRNA and protein expressions of iNOS and the levels of ALT and AST were all obviously decreased(P0.05or P0.01),and the edema and necrosis of hepatocytes were attenuated in group C.There were no significant differences in mRNA and protein expressions of eNOS between group B and group C(P0.05). Conclusion The rHuEPO may decrease the hepatic I-R injury via inhibiting the expression of iNOS in rats.
