川芎嗪对D-半乳糖损伤模型小鼠脑自由基代谢的影响 被引量:5

摘要 目的探讨川芎嗪对D-半乳糖(D-gal)损伤模型脑皮层和海马自由基代谢的影响。方法 C57BL/6J小鼠颈背部皮下注射2%D-gal100 mg.kg-1.d-1,连续40 d制造模型。第3周起各组分别用生理盐水(正常组、模型组)、川芎嗪(高、低剂量组)和哈伯因(阳性对照组)灌胃。第6周起进行Morris水迷宫测试,连续5 d。测试结束后,进行脑皮层、海马超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和Na+-K+ATP酶活力及丙二醛(MDA)含量的测定。结果与正常组相比,D-gal模型组学习记忆能力、皮层SOD、皮层和海马的GSH-Px和Na+-K+ATP酶活性均明显下降(P<0.05,P<0.001),海马SOD活性明显增高(P<0.005),皮层和海马MDA含量明显增高(P<0.05)。给予川芎嗪可明显提高模型小鼠的学习记忆能力(P<0.005,P<0.001),增强皮层SOD及皮层和海马的GSH-Px和Na+-K+ATP酶活性(P<0.05,P<0.001),降低海马SOD活性(P<0.05)及皮层和海马的MDA含量(P<0.001)。结论川芎嗪可明显提高模型小鼠的学习记忆功能,其作用机制与增强抗氧化活性有关。
