摘要
目的探讨小檗胺(berbamine,BER)对急性高眼压下视网膜损伤的保护作用及其机制。方法 96只大耳白兔随机分6组:正常对照组,阴性对照组,阳性对照组(VER),BER A、B、C组,后5组建立兔缺血-再灌注损伤模型,术后分时段观察视网膜形态、厚度的变化,并测量视网膜细胞内Ca2+含量的变化。结果阳性对照组,BER A、B、C组视网膜早期水肿较轻,晚期视网膜内层厚度明显厚于阴性对照组,术后168h BER C组视网膜内层厚度为(104.82±0.87)μm、阳性对照组为(105.14±0.92)μm,与正常对照组(105.94±0.66)μm相比,差异均无统计学意义(均为P>0.05)。造模后0h视网膜细胞中Ca2+含量开始升高,随再灌注时间延长呈逐渐增高趋势。BER组随再灌注时间的延长及药物浓度的增加,细胞内Ca2+含量逐渐减少,至术后168h,BER C组Ca2+含量为(45.35±0.48)μg·g-1,与正常对照组(44.61±0.84)μg·g-1相比,差异无统计学意义(P>0.05)。结论视网膜细胞内Ca2+超载,参与了视网膜缺血-再灌注损伤,BER通过抑制Ca2+内流从而保护视网膜细胞,并呈浓度依赖性。
Objective To study the protective effect of berbamine(BER) on retinal injury in rabbit with acute high intraocular pressure and its mechanisms.Methods Ninety-six New Zealand white rabbits were divided randomly into 6 groups:normal control group,negative control group,positive control group(VER),BER A group,BER B group,BER C group.The latter five groups established ischemia-reperfusion injury model of rabbits,the postoperative retinal morphous and thickness were observed at different time points,and the content of calcium ion(Ca2+) in retinal cells was detected.Results Positive control group,BER A,B,C group had less retinal edema in early stage and their retinal inner layer thickness in later stage were significantly thicker than that in negative control group,the retinal inner layer thickness at postoperative 168 hours was(104.82±0.87)μm in BER C group,(105.14±0.92)μm in positive control group,compared with the normal control group(105.94±0.66)μm,the difference was not statistically significant(both P0.05).The content of Ca2+ in rabbit retinal cells increased after immediate time of operation in treated groups,further increased with time dependent.With the dose of BER increasing and time prolonging,the content of Ca2+ decreased obviously till to normal level,which at postoperative 168 hours in BER C group was(45.35±0.48)μg·g-1,compared with normal control group(44.61±0.84)μg·g-1,there was no significant difference(P0.05).Conclusion Ca2+ overload in retinal cells may participate the ischemia-reperfusion injury,BER protect retinal cells by inhibiting Ca2+ influx in retinal cells with dose dependent.
