摘要
目的研究类风湿关节炎(RA)患者血清及外周血单核细胞中DcR3的表达情况。方法选取已确诊的60例类风湿因子(RF)(+)的RA患者并按照DAS28评分将其分为RAa组和RAb组,以30名体检健康者作为对照,酶联免疫吸附试验(ELISA)法检测各组血清可溶性陷阱因子3(DcR3)的表达情况。同时荧光实时定量聚合酶链反应(PCR)检测各组外周血单核细胞中DcR3的表达情况。采用t检验和,检验进行统计学分析。结果DcR3血清表达水平在DAS28〉2.6的RAb组为(264±72)ng/ml较健康对照组为(48±39)ng/ml明显升高(r=0.251,P〈0.05)。而在DAS28〈2.6的RAa组则没有差异。DcR3基因的扩增倍数在RAb组为23.5±5.4,健康对照组为8.3±3.6,差异具有统计学意义(r=0.336,P〈0.05)。结论DcR3在RA患者血清及外周血单核细胞中表达增高,而其血清表达水平在风湿活动较高的患者则更加明显。
Objective To study the decoy receptor 3 (DcR3) protein in the serum and its gene in PBMC from patients with rheumatoid arthritis (RA) and compared with healthy volunteers. Methods Sixty patients with rheumatoid arthritis whose RF were all positive, were grouped into RAa and RAb based on DSA28 scores. And 30 healthy volunteers were prepared. The expression of soluble DeR3 protein in the serum of RA patients and healthy volunteers was examined by enzyme-linked immunosorbent assay (ELISA). The expression of DcR3 gene in the PBMC of RA patients and healthy volunteers was examined by Real-time-PCR. Results The concentration of soluble DcR3 protein in serum of RAa group was (264±72) ng/ml and that of the control was (48±39) ng/ml. The expression of DcR3 in the sera of RA patients was significantly higher than that of healthy volunteers (r=0.251, P〈0.05). And a correlation between serum DcR3 concentration and the disease activities was seemed to be observed. The amplification altitude of the DcR3 gene of PBMC of RA group was 23.5±5.4 and the control was 8.3±3.6. The difference was significant (r=0.336, P〈0.05). Conclusion The DcR3 is highly expressed in patients with RA. And the seems DcR3 level is higher in patients with DAS28 grade 〉2.6 group.
