摘要 目的探讨Slit-Robo信号对粘液表皮样癌细胞系Mc3细胞生长与增殖作用的影响及其作用机制。方法免疫组织化学方法及流式细胞术细胞内因子检测法检测Mc3细胞Slit2及Robo1蛋白的表达;在Robo1受体胞外区的单克隆抗体R5作用下,以细胞计数法与软琼脂克隆形成实验观察Mc3细胞生长变化及用流式细胞术细胞内因子检测法检测增殖相关蛋白PCNA的表达。结果 Mc3细胞表达Slit2及Robo1蛋白,并且在R5单克隆抗体作用下,Mc3细胞体外生长能力受到抑制,增殖能力明显减弱,PCNA蛋白表达水平下降。结论 Slit-Robo信号可能影响Mc3细胞的生长增殖能力,对肿瘤细胞内的PCNA有正调控作用,提示该信号可能参与粘液表皮样癌的发生及发展过程。 Objective To study the role of Slit-Robo signaling in the proliferation of human mucoepidermoid carcinoma Mc3 cells.Methods We measured the expressions of Slit2 and Robo1 proteins in human mucoepidermoid carcinoma Mc3 cells using immunohistochemistry and flow cytometry.To test the role of Slit-Robo signaling in the proliferation of the cells,we treated the cells with the monoclonal antibody R5 against Robo1 receptor extracellular domain and observed the changes in the cell proliferation by cell counting and colonogenic assay;the expression of proliferating cell nuclear antigen(PCNA) in the cells following the treatment was measured with flow cytometry.Results Treatment of Mc3 cells with the monoclonal antibody R5 caused significantly suppressed cell growth and proliferation and obviously lowered the expression of PCNA.Conclusion Slit-Robo signaling can suppress the proliferation of Mc3 cells possibly by up-regulating the expression of PCNA.
