用酶解染色压片法观察水仙受精

摘要 运用酶解压片法结合整体染色,对水仙授粉后胚囊进行了分离和观察。经过用卡诺氏固定子房,系列酒精入水,25℃酶解2h,苏木精整体染色24h后取胚珠制片。压片时采用平移盖片加力方法,分离出不带任何珠心组织的完整胚囊。分离出来的胚囊可随盖片的平移而滚动,从而有利于从不同的角度进行观察。运用酶解染色压片技术,可以简便快速获得胚囊中受精作用的制片,值得进一步研究和应用。 By the method of enzyme-dye-squash technique, the Narcissus embryo sacs pollinated were isolated and observed. Ovaries were fixed in Carnoy's for 24 hours. After passing through a series of alcohol and distilled water, the ovaries were treated in 2% driselase aqueous solution for 2 hours at 25℃, and then dyed by hematoxylin for 24 hours. One ovule was taken to a slide with a drop of lacto-phenol-glycerin fruid and covered with a cover glass. Complete embryo sac without any nucellar tissue was isolated by the aid of parallel moving of the cover glass instead of tapping and pressing the cover glass. Isolated embryo sac can be rolled with the parallel moving of the cover glass to off er different observational angles. The process of fertilization in the embryo sac also was observed successfully by this technique. The enzyme-dye-squash technique is rather simple and is suitable not only to the research on embryo sac development, but also to the process of fertilization. The applicable value of the enzyme-dye-squash technique is discussed.
