T-bet、INF-γ mRNA在多发性肌炎/皮肌炎患者PBMC中的表达研究

Expression of T-bet and IFN-γ mRNA in PBMC of Patients with Polymyositis/dermatomyositis

[目的]探讨T-bet及 Th1细胞因子INF-γ与多发性肌炎（PM）/皮肌炎（DM）的关系.[方法]用RT-PCR方法检测多发性肌炎/皮肌炎（PM/DM）组及正常对照组外周血单个核细胞（PBMC）中T-bet、INF-γ的mRAN表达.[结果]PM组、DM组T-bet、INF-γ mRNA的表达强度均高于正常对照组,且两两组相比差异有显著性（P〈0.05）.[结论]T-bet表达增强可能与PM、DM的Th1细胞过度分化及细胞免疫增强有关.

[Objective]To explore the correlation of T-bet and Thl cytokine INF-γ with polymyositis （PM）/dermatomyositis（DM） patients. [Methods]The expression of T-bet and INF-γ mRNA in peripheral blood mononuclear cell （PBMC） of PM/DM patients and normal controls were measured by reverse transcriptase polymerase chain reaction（RT-PCR）. [Results] The expression intensity of T-bet and INF-γ mRNA in PM/DM patients were higher than those in normal controls, and there were significant differences between two groups（ P 〈0.05）. [Conclusion]The increased expression of T-bet mRNA may be correlated with overdifferentiation of Th1 cell and cellular immunity enhancement in PM/DM patients.
