摘要 目的:建立以高效液相色谱法测定异长春花碱脂质体中主药含量的方法。方法:色谱柱为Kromasil C1(84.6 mm×250 mm,5μm),流动相为乙腈-0.06 mol.L-1磷酸二氢钾缓冲液(pH3.0)(35︰65),流速为1.0 mL.min-1,检测波长为215 nm,柱温为30℃。结果:异长春花碱检测浓度的线性范围为2.08~20.8μg.mL-(1r=0.9999)。平均加样回收率为98.64%(RSD=1.09%)。结论:本方法简便、准确、重现性好,可用于该制剂的含量测定。 The study is aimed to establish a method to determine the content of Vinorelbine in liposomes by HPLC.The experiment was carried out on Kromasil C18(4.6 mm×250 mm,5 μm) column with the mobile phase consisted of acetonitrile-0.06 mol·L-1 KDP buffer(pH 3.0,35∶65) at a flow rate of 1.0 mL·min-1.The detection wavelength was at 215 nm and the column temperature was at 30℃.The result showed that the linear range of Vinorelbine was from 2.08 μg·mL-1 to 20.8 μg·mL-1(r=0.9999).The average recovery rate was 98.64%(RSD =1.09%).It is concluded that the method used in this analysis is simple,precise and replicable with high recovery and accuracy.It can be used to determine the content of total Vinorelbine in liposomes.
