摘要
在云南本地芥菜型油菜文山大脚黄(母本)与甘蓝型油菜品种花油5号(父本)杂交F1小孢子培养获得的DH群体中分离出双主茎材料,其田间表现具有良好的抗旱性。利用cDNA-AFLP技术分析七叶期甘蓝型油菜双主茎材料在干旱和对照条件下基因差异表达情况。共筛选了512对引物组合,获得56个干旱诱导上调表达的差异片段。对36个大于100 bp的片段克隆测序、同源比对、结果归并为30个非重复的TDFs,对已知功能的18个TDFs功能预测表明这些片段参与了细胞形态建成与降解、代谢、能量、细胞周期、蛋白质合成、蛋白质修饰、蛋白质结合、运输途径等相关过程。本试验获得的差异表达基因对于了解油菜苗期抗旱性形成的调控机制具有一定的参考价值。
The double caulis rapeseed was discovered from a DH population drived from F1 of Wenshandajiaohuang(Brassica juncea L.)×Huayou 5(Brassica napus L.).It showed strong drought resistance in the field.cDNA-AFLP was used to analyze genes differentially expressed under the condition of normal watering and serious drought in double caulis rapeseed(Brassica napus L.)seeding.Total 512 pair of AFLP primers were used to analyze expression profile.56 up-regulated transcript-derived fragments(TDFs) were acquired under drough stress.Among them,36 TDFs larger than 100 bps were sequenced.Gene function analysis revealed that most of TDFs were related with metabolism processes of cell formation and degradation,protein synthesis,processing and transporting,and energy metabolism,involving almost key metabolism processes during seedling development.This research could give basic information about the genes up-regulated respond to drought stress in Brassica napus for further studies of drought-resistance.
