摘要
以鸡腿168菌株为材料,采用Promoter Signal Scan软件对扩增序列进行启动子分析,Blastn同源性分析近缘真菌gpd启动子序列,并用MEGA4软件构建系统发育树。结果表明,该序列具有TAT-box和CAAT-box特征,且与GenBank中多个真菌gpd启动子序列同源性较高,基本证实该序列为鸡腿168 gpd启动子,可应用于鸡腿菇外源基因表达研究。
Promoter Signal Scan Software was used to analyze the promoter sequence of Coprinus comatus and gpd promoter sequence of its related species was operated Blastn homology analysis.The phylogenetic tree was constructed by MEGA4 Software.The results showed that the gpd promoter sequence had characteristics of TAT-box and CAAT-box and the promoter region of Coprinus comatus had high homology with that of some fungi in GenBank.It confirmed that the sequence was the 168 gpd promoter of Coprinus comatus and could be used to study foreign gene expression of Coprinus comatus.
