摘要
为了建立酶法提取老鹰茶中总黄酮的方法表是,将老鹰茶样品经一定量的纤维素酶于不同酶解温度、pH、酶解时间酶解,结合95%乙醇提取黄酮,分光光度法检测。结果:在酶浓度为0.40 mg/mL,酶解温度为50℃,pH 5.0,酶解时间为120 min的最佳酶解条件下,老鹰茶中黄酮的含量达到2.68%。表明,方法能满足茶叶中黄酮类化合物的提取要求。
To establish an enzymatic extraction method for extracting total flavonoids in Litsea coreana,Litsea coreana samples were extracted by cellulase enzyme of a specified level at different temperature,pH and the time of enzymatic hydrolysis.Combining of 95 % alcohol,flavonoids in Litsea coreana samples was extracted and determined by spectrophotometry.The rssults showed the optimum conditions for enzymatic hydrolysis were defined,which enzymatic hydrolysis temperature was 50 ℃,pH of enzyme solution was 5.0,the time of enzymatic hydrolysis was 120 min and the concentration of cellulase enzyme was 0.40 mg/mL.The content of flavonoids in the extraction liquid was as high as 2.68%.Therefore,this method was suitable for the extraction of flavonoids in teas.
