摘要
建立了利用钛胶高效液相色谱法测定食品中叶酸含量的方法。色谱条件:钛胶固定相(250mm×4.6mmi.d.,5μm),流动相为水:甲醇=70:30(v/v)、流动相中含磷酸盐(PBS)20mmol/L,流速0.9mL/min,柱温60℃,检测波长280nm。线性范围为5~25μg/mL,检出限1.9220μg/mL,回收率为92.7%~95.1%,RSD(n=8)<0.68%。应用于食品样品中叶酸的测定,结果满意。
A method for the determination of folic acid in food by high performance liquid chromatography on titania has been reported. The pretreated samples were separated with a titania-based column (250×4.6 mm, 5 um). The chromatographic separation conditions were as follows: a mixture of water and methanol in the ratio of 70 to 30 containing 20 m mol/L phosphate used as mobile phase, flow rate of 0.9 mL/ min, column temperature 60 ℃, and the detection wavelength 280 nm. The linear range of the method was 5-25 ug/mL with a detection limit of 1.92 ug/mL. The recoveries were between 92.7% and 95.1%. RSD(n=8) was less than 0.68%. The method is applied to determine folic acid in food with satisfactory results.
