摘要
目的观察塞来昔布对lewis肺癌小鼠血清galectin-3、MMP-9表达的影响。方法 C57BL/6小鼠45只随机分为正常对照组、荷瘤对照组和塞来昔布组。荷瘤对照组和塞来昔布组建立lewis肺癌模型。正常对照组和荷瘤对照组给予普通饲料喂养,而塞来昔布组自接种之日起给予含塞来昔布1 mg/g的饲料喂养。35天后测量各组皮下肿瘤的重量和肺部肿瘤转移灶的数目。并且使用ELISA检测小鼠血清MMP-9和galectin-3水平。结果塞来昔布组皮下肿瘤重量和肺部肿瘤转移灶数量明显低于荷瘤对照组(P<0.05);与荷瘤对照组相比,塞来昔布组血清MMP-9和galectin-3的水平明显降低(P<0.05)。结论塞来昔布能够抑制lewis肺癌的生长和转移,其机制可能与调节血清galectin-3、MMP-9的表达有关。
Objective To study the effect of eelecoxib on the serum galcetin-3 level in lewis lung carcinomas hearing mouse. Methods Forty-five C57BL/6 mice were divided randomly into three groups: the control group, tumor-bearing control group and celeeox- ib group. Lewis lung carcinoma models were established in the tumor-bearing control group and eeleeoxib group. Cclecoxib group were given the diet with eeleeoxib 1 mg/g on the day after implantation throughout the experiment, whereas the control group and the tumor-bearing group were given normal food. Weights of tumor and lung metastasis were measured 35 days after tumor cell inoculation. Enzyme-linked immunosorbent assay was performed to determine the serum galectin-3 and MMP-9 level. Result The weights of tumor in the eelecoxib group was significantly smaller than that in the tumor-bearing group ( P 〈 0. 05 ). The number of lung metastasis in the celeeoxib group was significantly smaller than that in the tumor-bearing group ( P 〈 0.05 ). The serum galectin-3 level and MMP-9 level in the tumor-bearing control group were significantly higher than that in the ecleeoxib group ( P 〈 0. 05 ). Conclusion Celecoxib can inhibit the growth and decrease the number of lung metastatic tumors in the mice, which was probably because that eelecoxib modulate the expression of galectin- 3 of lewis lung carcinoma.
