重楼总皂苷对HaCaT细胞增殖和分泌IL-8的影响

Effect of Steroidal Saponins from P.polyphylla on Proliferation of HaCaT Cells and Secretion of IL-8

目的研究重楼总皂苷对人永生化角质形成细胞株(HaCaT细胞)增殖和分泌白介素8(IL-8)的影响。方法采用MTT法进行细胞增殖实验,采用ELISA法测定重楼总皂苷对HaCaT细胞分泌IL-8的情况。结果在所选浓度范围内(60μg/mL,90μg/mL,120μg/mL和150μg/mL),重楼总皂苷对HaCaT细胞的增殖有抑制作用;其中120μg/mL和150μg/mL浓度组重楼总皂苷可抑制HaCaT细胞自分泌IL-8,90μg/mL,120μg/mL和150μg/mL浓度组重楼总皂苷能抑制TNF-α(浓度为100U/mL),刺激HaCaT细胞分泌IL-8。结论重楼总皂苷能抑制HaCaT细胞增殖和IL-8的分泌。

Objective To study the effect of steroidal saponins from P. polyphylla on proliferation of HaCaT cells and secretion of IL-8. Methods The inhibition rate of steroidal saponins from P. polyphylla for proliferation of HaCaT cells was detected by MTT. The influence of steroidal saponins from P. polyphylla on secretion of IL- 8 was determined by ELISA. Results Within the selected concentration （60μg/mL, 90μg/mL, 120μg/ mL, 150 μg/mL）, steroidal saponins from P. polyphylla inhibited the proliferation of HaCaT cells compared with control groups. The spontaneous secretion of IL-8 from HaCaT cells was decreased after treatment with steroidal saponins （120μg/mL, 150μg/mL）, meanwhile, The secretion of IL-8 from HaCaT cells by TNF-α stimulation was inhibited after treatment with steroidal saponins （90μg/mL, 120μg/mL, 150 μg /mL）. Conclusion Steroidal saponins from P. polyphylla can suppress the growth of HaCaT cells and inhibite se- cretion of IL-8 from HaCaT cells.
