B-cell-specific Moloney murine leukemia virus integration site-1 expression in gastric carcinoma and its clinical significance

摘要 目的:探讨B细胞特异性莫洛尼鼠白血病病毒插入位点-1(B-cell-specific Moloney murine leukemia virus integration site-1,Bmi-1)蛋白在胃癌组织中的表达及其临床意义。方法:应用免疫组织化学法检测正常胃黏膜、慢性非萎缩性胃炎、慢性萎缩性胃炎、异型增生及胃癌组织中Bmi-1、c-myc和Zeste基因增强子同源物2(enhancer of Zeste homolog 2,EZH2)蛋白的表达。结果:Bmi-1蛋白在正常胃黏膜、慢性非萎缩性胃炎、慢性萎缩性胃炎、异型增生和胃癌组织中的阳性表达率分别为5.0%、16.7%、30.7%、58.1%和78.0%,各组间差异有统计学意义(P<0.05)。胃癌组织中Bmi-1高表达率为45.0%,其表达水平与肿瘤大小、分化程度、浸润深度、淋巴结转移和TNM分期有关(P<0.05)。胃癌组织中Bmi-1的表达与c-myc、EZH2表达之间呈正相关(rs=0.567,P<0.001;rs=0.582,P<0.001)。结论:Bmi-1在胃癌组织中表达上调,其高表达与胃癌的分化程度、浸润深度和淋巴结转移有关。 Objective: To investigate the expression of B-cell-specific Moloney murine leukemia virus integration site-1 (Bmi-1) in gastric carcinoma and its clinical significance. Methods: The expressions of Bmi-1, c-myc, and enhancer of Zeste homolog 2 (EZH2) in normal gastric mucosa, chronic non-atrophic gastritis, chronic atrophic gastritis, dysplasia and gastric adenocarcinoma tissues were detected by immunohistochemistry. Results: The positive expression rates of Bmi-1 in normal gastric mucosa, chronic non-atrophic gastritis, chronic atrophic gastritis, dysplasia and gastric carcinoma tissues were 5.0%, 16.7%, 30.7%, 58.1% and 78.0%, respectively. There was a significant difference in positive expression rate of Bmi-1 among different gastric tissues (P〈O.05). The high expression rate of Bmi-1 (45.0%) in gastric adenocarcinoma tissues was associated with tumor size, differentiation, invasion depth, lymph node metastasis and TNM stage (P〈0.05). The Bmi-1 expression was positively correlated with the expressions of c-myc (rs = 0.567, P〈0.001) and EZH2 (rs = 0.582, P〈0.001). Conclusion: The expression level of Bmi-1 is up-regulated in gastric carcinoma, and which is related with differentiation, invasion depth and lymph node metastasis.
