摘要
目的研究高氧对体外培养的人胚肺成纤维细胞(HELF)的毒性作用。方法将生长良好的HELF传代后,用无血清培养基培养24h,分别置于空气及体积分数90%O2的细胞培养箱中培养5d,采用噻唑蓝(MTT)法检测细胞活力,化学比色法检测细胞上清液中乳酸脱氢酶(LDH)及超氧化物歧化酶(SOD)的活性。结果与空气对照组相比,在给予体积分数90%O22d时细胞上清液中LDH含量明显升高,SOD含量则明显减低(P<0.05),且随高氧暴露时间延长,呈现出明显的时间依赖关系(3~5d,P<0.01),细胞大量凋亡。结论体积分数90%O2对体外培养的HELF具有明显的细胞毒性作用,直接引起细胞损伤,加速其凋亡。
Objective To explore the cytotoxieity of hyperoxia on human embryo lung fibroblasts (HELF) in vitro. Methods After HELFs were passaged and cultured 24 hours in DMEM with no fetal bovine serum, they were then randomly divided into air and 90% O2 group,then cultured for 5 days. The vigor of HELFs was assessed by MTT assay, lactate dehydrogenase(LDH) and superoxide dismutase(SOD) activity in the supernatant of the culture medium were measured by chemical colorimetry. Results As compared with air group, after exposure to 90 % O2 2 days, the LDH activity significantly increased and the SOD activity significantly decreased(2 d,P〈0. 05; 3-5 d,P〈0.01), caused an increased cell apoptosis of HELFs. Conclusion 90% O2 is a concentration that caused severe cell damage and lead to cell apoptosis in vitro .
