胃癌组织中miR-181a和miR-27a高表达及意义 被引量:4

摘要 目的:分析miR-181a与miR-27a两种miRNAs在胃癌中的表达及其与胃癌临床病理特征的关系。方法:选取广州市第一人民医院胃癌外科手术切除的胃癌标本50例和对应的距离胃癌病灶5 cm以上的癌旁组织,以及25例慢性浅表性胃炎黏膜为对照,采用实时荧光定量-PCR(qRT-PCR)对miR-181a和miR-27a作定量分析。结果:相对于自身癌旁组织和他人的慢性浅表性胃炎组织,miR-181a和miR-27a在胃癌组织中表达上调,差异有统计学意义(P<0.05)。胃癌者的性别、年龄、病例分型和淋巴结转移与miR-181a和miR-27a表达无显著相关性。结论:miR-181a和miR-27a在胃癌组织中显著表达,可能参与胃癌的发生过程。 Objective:To investigate expression levels of the miR-181a and miR-27a in gastric cancers, and their correlation with clinicopathological features. Methods : Fifty pairs of surgically resected specimens of gastric carcinoma and adjacent tissue (at least 5cm from the lesions) were obtained. Another 25 mucosa samples from patients with chronic superficial gastritis were included as controls. Quantitative real-time PCR analysis was performed to detect the expressions of miR-181a and miR-27a in these samples. Results:The miR-181a and miR-27a expression in gastric cancers was up-regulated compared with adjacent non-cancerous tissues and mucosa in chronic superficial gastritis (both P 〈 0.05 ). Both miR-181a and miR-27a expressions were not statistically associated with patient age, gender, pathology and lymph node metastasis. Conclusion: Both miR-181a and miR-27a are over-expressed in gastric carcinoma tissues and may play a role in development of the disease.
