摘要
目的:建立测定人血清中舒芬太尼浓度的简便方法。方法:血清中舒芬太尼用乙酸乙酯-石油醚(4:1)单步提取后采用反相高效液相色谱法进行测定,其中色谱柱为Symmetry C18,流动相为乙腈-0.02mol.L-1醋酸铵-盐酸(pH4.3)(45:55),检测波长为230nm,柱温为室温,内标为艾司唑仑。结果:舒芬太尼血药浓度在0.025~4.000μg.mL-1范围内线性关系良好(r=0.9999);日内、日间RSD分别为3.49%~4.51%、4.67%~5.99%,平均回收率为101.92%~107.04%。结论:本方法提取步骤少,方法简单、快速、专一性强、准确,适合人血清中舒芬太尼的浓度监测。
OBJECTIVE: To establish the method for the determination of sufentanil concentration in human serum. METH- ODS: Sufentanil was extracted from serum by ethyl ethanoate-petroleum ether(4:1) and determined by RP-HPLC. The determination was performed on Symmetry C18 column with mobile phase consisted of acetonitrile-0.02 mol. L ^-1 NH4Ac-HCL (pH4.3)(45: 55). The detection wavelength was set at 230 nm and the column temperature was room temperature. Estazolam was used as internal standard. RESULTS: The linear range of sufentanil was 0.025-4.000μg·mL^-1(r=0.999 9) with intra-day and inter-day RSDs of 3.49% -4.51% and 4.67% -5.99%. Average recoveries were 101.92% -107.04%. CONCLUSION: The method is simple, rapid, accurate and specific with few steps. It is suitable for drug concentration monitoring of sufentanil in human serum.
