干扰素-α联合柴胡皂苷对免疫性肝损伤小鼠血清T淋巴细胞亚群和肝组织细胞因子的影响 被引量:15

摘要 目的观察干扰素-α联合柴胡皂苷对免疫性肝损伤小鼠血清T淋巴细胞亚群和肝组织细胞因子水平的影响。方法小鼠随机分为正常对照组(正常组)、模型组、干扰素组、柴胡皂苷组、联合组(干扰素+柴胡皂苷)5组,各组分别给药2天并注射ConA后8、24h采集标本,检测外周血淋巴细胞亚群,肝组织肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-18(interleukin-18,IL-18)、白细胞介素-10(in-terleukin-10,IL-10)水平,血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性和观察肝脏组织病理改变。结果 (1)与正常组比较,模型组肝组织炎症坏死显著,血清ALT、AST明显升高,同时24h外周血CD4+及CD8+T淋巴细胞比例和肝组织IL-10水平明显降低(P<0.01),肝组织IL-18、TNF-α水平显著升高(P<0.05,P<0.01)。(2)与模型组比较,3个用药组肝组织散见点、片状坏死,血清ALT、AST活性显著降低(P<0.01),同时外周血CD4+及CD8+T淋巴细胞比例和肝组织IL-10水平明显升高(P<0.01,P<0.05),而IL-18、TNF-α水平显著降低(P<0.05,P<0.01)。(3)联合组24hCD4+T淋巴细胞和CD8+T淋巴细胞比例较干扰素组明显升高,8h和24h的IL-18水平较干扰素组明显降低,24hTNF-α水平较干扰素-α组显著降低(P<0.05)。结论干扰素联合柴胡皂苷能有效发挥抗免疫性肝损伤作用。 Objective To observe the effects of interferon-a combined with saikosaponin on serum T lymphocyte subgroups and hepatic cytokines in mice with immune hepatic injury. Methods The mice were randomly divided into five groups, i.e., the normal control group, the model group, the interferon group, the saikosaponin group, and the interferon combined saikosaponin group. Corresponding medication was given to mice in respective groups for two days. Peripheral blood was collected 8 and 24 h after concanavalin A (Con A) was injected. Serum lymphocyte subgroups, tumor necrosis factor a (TNF-a), levels of interleukin 18 (IL-18) and IL-10, activities of alanine aminotransferase (ALT) and aspartate transaminase (AST) were detected. Pathological changes of the liver tissue were observed. Results (1) Compared with the normal control group, serious inflammation and necrosis was significant in the liver tissue of the model group. The serum levels of ALT and AST obvious- ly increased. Meanwhile, the 24-h peripheral blood CD4 T cell and CD8+ T cell ratios and the hepatic IL-10 level obviously decreased (P 〈 0.01 ). The levels of IL-18 and TNF-a significantly increased (P 〈 0. 05, P 〈 0.01 ). (2) Compared with the model group, dot and lamellar necrosis was dispersedly seen in the liver tissue in the three medication groups. The serum activities of ALT and AST significantly decreased ( P 〈 0.01 ). Meanwhile, the peripheral blood CD4+ T cell and CD8+ T cell ratios, as well as the hepatic IL-10 level obviously increased ( P〈 0.01, P〈0.05). The levels of IL-18 and TNF-a significantly decreased ( P〈0.05, P〈0. 01 ). (3) In the interferon combined saikosaponin group, the 24-h peripheral blood CD4 T cell and CD8+ T cell ratios increased more obviously than those of the interferon group. The 8- and 24-h IL-18 levels were obviously lower than those of the interferon group, while the 24-h TNF-~ level significantly decreased more than that of the interferon group ( P 〈 0. 05). Conclusion Interferon-a combined saikosapon could effectively play a role in fighting against the immunehepatic injury.
