摘要
目的观察姜黄素(Curcumin)对人骨肉瘤MG63细胞增殖及细胞内Wnt/β-catenin信号通路相关基因表达的影响,探讨姜黄素抑制MG63细胞生长的机制。方法体外培养MG63细胞,用不同浓度的姜黄素处理细胞不同时间,MTT法检测MG63细胞的增殖活力;RT-PCR法检测细胞内β-catenin和CyclinD1基因mRNA的转录水平;Western blot检测β-catenin和CyclinD1蛋白的表达。结果姜黄素可抑制MG63细胞的增殖,其抑制作用呈剂量-时间依赖性,当姜黄素浓度为20.00μmol/L时,对细胞增殖的抑制作用最明显;姜黄素可显著降低细胞内β-catenin和CyclinD1基因mRNA的转录水平和蛋白的表达水平,且呈剂量-时间依赖性。结论姜黄素可抑制MG63细胞增殖,其机制可能是通过抑制Wnt/β-catenin信号通路的活性实现的,这将为临床治疗骨肉瘤提供新的思路。
Objective To observe the effect of eureumin on proliferation of human osteosareoma MG63 cells and expression of Wnt/ β-catenin signaling pathway-associated gene, and investigate the mechanism of inhibiting the growth of MG63 cells by curcumin. Methods MG63 cells were cultured in vitro, and treated with eureumin at various concentrations for various hours, then determined for proliferation activity by MTr method, for transcription levels of β-eatenin and CyciinD1 mRNAs by RT-PCR, and for expressions of ^-catenin and CyclinD1 by Western blot. Results Cureumin showed dose- and time-dependent inhibitory effect on the proliferation of MG63 cells. However, the inhibitory effect of curcumin at a concentration of 20. 00 p, mol/L was superior to those at other concentrations. Meanwhile, eurcumin decreased the mRNA transcription and protein expression levels of β-catenin and CyclinD1 in dose- and time-dependent modes. Conclusion Curcumin inhibited the proliferation of MG63 cells by a possible mechanism of inhibiting the activity of Wnt/ ^-eatenin signaling pathway, which provided a novel route for clinical therapy of osteosarcoma.
