摘要
目的通过研究大鼠急性脊髓损伤(SCI)后血清β-淀粉样前体蛋白(B-APP)、超敏C反应蛋白(hs-CRP)不同时间点含量变化,探讨其在继发性SCI中的作fH机制及其时程变化规律。方法选用健康成年雌性SD大鼠90只,采用改良Allen法制备大鼠脊髓完全损伤(完全损伤组,30只)和不全损伤(小完全损伤组,30只)动物模刹,另30只大鼠仅行椎板切除作为假手术组。分别于造模成功后12h、24h、3d、7d、14d采集股静脉血,ELISA检测不同程度SCI大鼠伤后血清B-APP和hs-CRP含量变化。结果B-APP于急性SCI后24h达到最高水平,hs-CRP第3天达到高峰。完令损伤组各时间点β-APP和hs.CRP水平分别与不拿损伤组和假手术组比较差异均有统计学意义(P〈0.05);不全损伤组与似乎术组除14dβ-APP和hs-CRP水平差异无统计学意义(P〉0.05)外,其余各时间点两组的p-APP、hs-CRP水平比较差异均有统计学意义(P〈0.05)。结论急性SCI大鼠血清p-APP、hs.CRP水平的变化反应了SCI的严重程度,β-APP较hs-CRP的变化敏感,住一定时限内可敏感地反映SCI的严重程度及预后。大鼠血清β-APP、hs-CRP的动态变化提示二者共同参与了急性继发性SCI。
Objective To investigate the serum levels of β-amyloid precursor protein (β-APP) and hypersensitive-cross-reactiwr protein (hs-CRP) after acute spinal cord injury (SCI) in rats at different time points and study their roles in secondary spinal cord injury. Methods Ninety healthy adtdt female SD rats were ramtomly assigned into 3 ewm groups. Improved Allen method was used to create models of complete SCI (group A) and models of incomplete SCI (group B). Ira group C only laminectomy was performed without SCI. The serum was collected at 12 hours, 24 hours, 3 days, 7 days, and 14 days after operation through femoral vein to detect levels of β-APP and hs-CRP by EIdSA. Results The serum level of β-APP reached the peak value 24 hours after acute SCI, and the serum lerum of hs-CRP reached the peak 3 days after acute SCI. There were significant differences between groups A and C ira β-APP and hs-CRP levels at all time points ( P 〈 0. 05) . There were also significant differences between groups B and C in β-APP and hs-CRP levels at all time points ( P 〈 0.05) except tit 14 days ( P 〉 0.05). Conclusions The serum lerum of β-APP and hs-CRP are positively associated with the severity of traumatic SCI. β-APP may be a more sensitive indicator than hs-CRP in a certain time. The dynamic changes of serum β-APP and hs-CRP suggest involve- merit of the 2 proteins in the secondary spinal cord injury.
