摘要
目的:本研究旨在通过小鼠胚胎发育时期涎腺miRNA基因芯片的检测,研究miRNA靶基因调控小鼠涎腺胚胎发育机制以及相互联系。方法:选择在胚胎发育第18天、第19天、及出生当天、出生后3d4个时期形态发育完整下颌下腺作为标本,通过检测基因芯片,建立基因表达谱,运用生物信息学方法,分析这4个时段miRNA表达及相互联系,靶基因的调控关系。结果:建立miRNA与靶基因间调控网络。结论:miRNA在调节小鼠细胞周期、脂质代谢、离子转运、细胞增殖过程中起重要作用,从而使样本在胚胎期到出生后样本发生表型上的变化。
Objective: To study the regulation of miRNAs chips of salivary gland to target gene and the relationship between them during embryonic development period in mice. Method: Submandibular glands of mice were dissected at selected times during development(El8.5, E19.5 .P0 .P3). Using miRNAchips analysis, miRNA expression pro- files had been established. Then the expression of the miRNA and the relationship between miRNAs and target gene were analyzed by bioinformation method. Results: The miRNA--Gene--Net is established. Conclusion: miRNAs play an important part in regulating cell cycle, lipid metabolic process, ion transport, cell proliferation, and etc. Then, the salivary gland phenotype changes from embryonic stage to birth in mice.
