赤芍总苷对人黑色素瘤细胞A375血管生成因子VEGF和Ang-2的影响 被引量:9

The effect of total paeonyglucosides on VEGF and Ang-2 expressions in human malignant melanoma cell

摘要 目的探讨赤芍总苷(TPG)对人黑色素瘤A375细胞血管内皮生长因子(VEGF)和促血管生成素-2(Ang-2)表达的影响。方法体外培养人黑色素瘤A375细胞,分别加入12.5、25、50、100、200mg/LTPG,对照组加入等量生理盐水,孵育24h、48h后,MTT方法测定生长抑制率;RT-PCR、Western blot分别测定TPG处理48h后VEGF和Ang-2 mRNA和蛋白的表达。结果 MTT结果显示TPG对黑色素瘤细胞增殖有显著抑制作用;RT-PCR、Western blot结果显示,TPG可显著抑制人黑色素瘤细胞VEGF、Ang-2 mRNA水平及蛋白水平的表达。结论 TPG具有显著的抗黑色素瘤瘤血管生成的作用,其机制可能与下调VEGF、Ang-2的表达有关。 Objective To explore the effects of total paeonyglucosides(TPG) on VEGF and Ang-2 expressions in human malignant melanoma A375 cell.Method Human malignant melanoma cells were cultured with TPG(12.5,25,50,100,200mg/L) for different time(24h or 48h) in vitro,and control group was cultured with NS.Growth suppression rates were tested by MTT.Cells treated with 25 mg/L TPG for 48h,and then using RT-PCR and Western blot for determining expression of VEGF and Ang-2 at mRNA and protein level.Results MTT results indicated that the proliferation of human malignant melanoma cells were inhibited by TPG of different concentrations.Compared with the control group,mRNA and protein levels of VEGF and Ang-2 were decreased in the TPG groups(all P0.05).Conclusions TPG had the effect of inhibiting vascularization by down-regulating the expressions of VEGF and Ang-2.
