生物表面活性剂发酵条件的响应面分析优化 被引量:1

摘要 本文利用Plackett-Burman设计法和响应面分析法,对产生物表面活性剂的采油菌株DN4-3的发酵条件进行优化,筛选出三个显著影响因子(葡萄糖用量、酵母浸粉用量和氯化钠用量)。在此基础上,通过最陡爬坡实验和响应面分析,研究了显著因素的最佳水平及其交互作用,确定了最佳发酵条件。最佳发酵条件为:葡萄糖27.83 g/L、酵母浸粉2.73 g/L、NaCl 2.37 g/L、KH2PO4 9 g/L、K2HPO4 3 g/L、CaCl2 0.16 g/L、MgSO4.7H2O 0.5 g/L、接种量4%、种龄16 h、装液量250 mL/500 mL、温度37℃、初始pH值7.0、摇床转速160 r/min。采用优化后条件进行摇瓶发酵,产表面活性剂1025.98 mg/L,与预测值很接近,比优化前提高了35.43%。 The fermentation conditions of biosurfactant production by strain DN4-3 were optimized with the Plackett-Burman design and response surface analysis.Plackett-Burman design was used to evaluate the influence of related factors,as a result,three significant factors,such as the content of glucose,the content of yeast extract power and NaCl content,were selected.The path of steepest ascent and response surface analysis were used to determine the optimal levels of the significant factors.Based on the single factor experiments,the optimal medium composition and fermentation conditions were determined as follows: glucose 27.83 g/L,yeast extract power 2.73 g/L,NaCl 2.37 g/L,KH2PO4 9 g/L,K2HPO4 3 g/L,CaCl2 0.16 g/L,MgSO4· 7H2O 0.5 g/L,inoculation amount 4%,seed age 16 h,medium volume 250 mL/500 mL,temperature 37℃,initial pH 7.0,rotation speed 160 r/min.Under the optimal conditions,1025.98 mg/L biosurfactant was produced by the strain DN4-3,which increased about 35.43% than before.
