多重等位基因特异性PCR-熔解曲线分析快速定型β珠蛋白基因突变 被引量:2

摘要 目的建立自动化、高通量、准确快速检测β珠蛋白基因突变的技术。方法在反应体系中加入新型荧光染料LC Green进行实时荧光聚合酶链反应,反应结束后,以0.1℃/s变化速度从60℃至95℃每隔3 s记录一次荧光值,获得熔解曲线,根据得到的熔解曲线和Tm值分析定型β地中海贫血的CD41-42、IVS 2nt654、CD17、CD71-72四种等位基因突变。结果根据Tm值能准确判断基因型,检测CD41-42、IVS 2nt654、CD17、CD71-72四种等位基因的Tm值分别为78±1℃、80±1℃、76±1℃、79±1℃。结论该技术具有自动化程度高、不需荧光标记探针、成本低、易质控、防污染、高通量等优点,适于临床推广应用。
