摘要
目的研究取样对精子浓度及活力分析结果的影响,并通过不同的方式进行混匀,以减小取样误差。方法分别进行两种实验,即实验一和实验二。实验1有55例,是对同一例精液标本,混匀后,随机取样4次:实验2有共47例,将其分为3组,组1是标本不进行混匀;组2是将标本上下颠倒混匀10次;组3是将标本用巴斯德吸管吹打混匀5次,分别在标本底部和顶部进行取样分析。对每次取样结果用HamiltonCASA系统进行精子浓度和活动力分析并记录。结果实验1:对同一标本的4次取样中,任意两次取样的精子浓度和活力之间的相关性比较低。实验2:(1)组1两次取样的浓度比较差异有统计学意义(P〈0.05),活动力比较差异无统计学意义(P〉0.05)。(2)组2两次取样的浓度和活动力之间均无显著性差异(P〉0.05);(3)组3两次取样的浓度和活动力之间比较差异均无统计学意义(P〉0.05)。结论对同一例标本,不同取样之间浓度和活动力的差异范围较大,相关性比较低。取样误差主要表现在精子浓度上,而精子活动力差异不大。所以,对精液标本,在取样前必须充分混匀,上下颠倒10次是一个比较简单有效的方式。
Objective To study the effect of sampling method on sperm concentration and sperm motility and find the best mixing ways to reduce sampling error. Methods There were two groups of experiment. For experiment 1(55 cases), sperm samples were mixed well and sampled randomly 4 times. For experiment 2, 47sperm cases were randomly divided into 3 groups including group 1 (sperm samples were not mixed), group2(semen samples were mixed by upside down I0 times) and group3 (semen samples were mixed with Pasteur pipettes). Sperm samples were sampled at the top and the bottom. Sperm concentration and motility were analyzed by CASA. Results For sperm samples from 4 times sampling in experiment 1, sperm concentration was lowly correlaed with sperm motility. There was a significant difference in sperm concentration but no significant difference in sperm motility in Groupl. There were no significant differences both in sperm concentration and motilityin Group2. There were no significant differences in sperm concen- tration and motility in Group3. Conclusion There were some obvious differences in sperm density and sperm motility between two sampling methods, which have a low correlation. The difference of sperm density was observable, but no sperm motility between different sampling methods,. Sperm samples had better be mixed well. Sperm samples mixing by upside down 10 times could be a simple and effcetive method for semen routine analysis.
