摘要
目的:研究针刺对更年期脑缺血大鼠缺血区脑组织GFAP和OX42蛋白表达的影响,探讨针刺对神经损伤的保护作用及可能机制。方法:雌性SD大鼠,经筛选为自然更年期,随机分为假手术组、缺血模型组和针刺组,采用右侧大脑中动脉阻塞(MCAO)法建立脑缺血动物模型。针刺治疗后,采用Ber-derson神经体征评分法,观察大鼠神经行为学变化,光镜观察脑组织的病理学变化;免疫组织化学法检测脑组织中GFAP和OX42蛋白表达。结果:针刺能明显改善大鼠脑缺血后24 h和7 d的神经损伤症状;改善脑组织病理形态学损伤状态;对缺血24 h和7 d脑组织中GFAP和OX42蛋白表达有不同程度降低,与模型组相应时间点相比较,差异有统计学意义(P﹤0.05或P﹤0.01)。结论:针刺能够抑制胶质细胞GFAP和OX42的活化,减轻脑缺血后的炎症级联反应,从而改善脑组织病理学损伤状态,起到神经保护作用。
Objective:To research the influence of GFAP and OX42 protein expression of MCAO climacteric female rats by electro-acupuncture,and investigate the protective effect of neurological damage and possible mechanisms. Methods:Healthy naturally climacteric female SD rats,use intraluminal MCAO rat model,after the electroacupuncture treatment,Experiments were carried out after the modeling 24 h and 7 d.Neurological signs by Bederson neurological score behavioral changes were observed;Use light microscope morphological changes in brain tissue;It was detected by immunohistochemistry in brain tissue GFAP,OX42 expression. Results:Electroacupuncture can be significantly improved after cerebral ischemia in rats 7 d or 24 h symptoms of nerve injury;It could improve the pathological brain damage state and reduce varying degrees of the protein expression of GFAP and OX42 in 7 d and 24 h ischemic brain tissue.Compared with model group,there were significant differences(P﹤0.05 or P﹤0.01). Conclusion:The electroacupuncture can significantly improve the state of brain tissue pathological damage,and improve after MCAO in rats 7 d or 24 h and symptoms of neurological damage.Electroacupuncture can be through activation Inhibition of glial cells the GFAP and OX42,24 h and 7 d after ischemia,and inhibit the post-ischemic 24 h and 7 d inflammatory cascade.Reduce the blood-brain barrier damage so as to improve brain histopathological damage.It plays a positive role in brain protection.
