活血化瘀法对慢性盆腔炎雌性大鼠血清TGF-β1和IL-4、IL-10水平的影响 被引量:37

Effect of promoting blood circulation by removing blood stasis on serum transforming growth factor-β1,interleukin-4 and interleukin-10 levels in female rats with chronic pelvic inflammatory disease

摘要 目的:探讨活血化瘀法对慢性盆腔炎雌性大鼠血清TGF-β1、IL-4、IL-10水平的影响。方法:采用混合细菌加机械损伤方法,建立慢性盆腔炎大鼠模型,对60只大鼠随机分为模型组、桂枝茯苓胶囊治疗高、中、低剂量组、对照组,各组均为12只,另外选择假手术慢性盆腔炎造模大鼠(假手术组)、正常饲养未干预大鼠(正常组)各12只。检测各组大鼠血清TGF-β1、IL-4、IL-10的水平。结果:①血清TGF-β1水平:模型组较正常组、假手术组显著升高(P<0.01);实验组较模型组显著降低(P<0.01或P<0.05),且中、高剂量组水平接近正常组(P>0.05);低剂量实验组较对照组显著升高(P<0.01);中剂量实验组较高、低剂量实验组均降低。②血清IL-4、IL-10水平:模型组较正常组、假手术组显著降低(P<0.01);中剂量实验组较模型组明显升高(P<0.01),且接近正常组水平(P>0.05);中、高剂量实验组较对照组均升高,其中IL-4升高有统计学意义(P<0.01),IL-10升高无统计学意义(P>0.05),低剂量实验组较对照组均降低,其中IL-4无统计学意义(P>0.05),IL-10有统计学意义(P<0.01);中剂量实验组较高、低剂量实验组均升高。③中剂量实验组各指标间相关性分析:TGF-β1与IL-4、IL-10有显著负相关性(P<0.01),IL-4与IL-10有显著正相关性(P<0.01)。结论:活血化瘀法能影响慢性盆腔炎大鼠血清TGF-β1、IL-4、IL-10水平,与药物剂量存在相关性,这可能是活血化瘀法抗粘连和抑制炎症反应,从而治疗慢性盆腔炎和缓解盆腔粘连的机制之一。 Objective:To explore the effect of promoting blood circulation by removing blood stasis on serum transforming growth factor-β1(TGF-β1),interleukin-4(IL-4) and interleukin-10(IL-10) levels in female rats with chronic pelvic inflammatory disease.Methods:Mixed bacteria plus mechanical damage method were used to establish rat model of chronic pelvic inflammatory disease,60 rats were divided into model group,high dose cassia tuckahoe capsule group,middle dose cassia tuckahoe capsule group,low dose cassia tuckahoe capsule group and control group randomly,12 rats in each group;12 rats in sham operation group and 12 rats in normal group were also selected.ELISA was used to detect the serum levels of TGF-β1,IL-4 and IL-10 in the seven groups.Results:①The serum level of TGF-β1 in model group was significantly higher than those in normal group and sham operation group(P0.01);the serum level of TGF-β1 in experimental groups was significantly lower than that in model group(P0.01 or P0.05),and the serum levels of TGF-β1 in high dose cassia tuckahoe capsule group and middle dose cassia tuckahoe capsule group were similar to that in normal group(P0.05);the serum level of TGF-β1 in low dose cassia tuckahoe capsule group was significantly higher than that in control group(P0.01);the serum level of TGF-β1 in middle dose cassia tuckahoe capsule group was significantly lower than that in high dose cassia tuckahoe capsule group and low dose cassia tuckahoe capsule group.②The serum levels of IL-4 and IL-10 in model group were significantly lower than those in normal group and sham operation group(P0.01);the serum levels of IL-4 and IL-10 in middle dose cassia tuckahoe capsule group were significantly higher than those in model group(P0.01),and the serum levels of IL-4 and IL-10 in middle dose cassia tuckahoe capsule group were similar to those in normal group(P0.05);the serum levels of IL-4 and IL-10 in middle dose cassia tuckahoe capsule group and high dose cassia tuckahoe capsule group were higher than those in control group,and there was significant difference in the serum level of IL-4(P0.01),while there was no significant difference in the serum level of IL-10(P0.05);the serum levels of IL-4 and IL-10 in low dose cassia tuckahoe capsule group were lower than those in control group,and there was no significant difference in the serum level of IL-4(P0.05),while there was significant difference in the serum level of IL-10(P0.01);the serum levels of IL-4 and IL-10 in middle dose cassia tuckahoe capsule group were significantly higher than those in high dose cassia tuckahoe capsule group and low dose cassia tuckahoe capsule group.③There was a significant negative correlation between serum TGF-β1 level and serum IL-4,IL-10 levels(P0.01),there was a significant positive correlation between serum IL-4 level and serum IL-10 level(P0.01).Conclusion:Promoting blood circulation by removing blood stasis can affect the serum levels of TGF-β1,IL-4 and IL-10 in female rats with chronic pelvic inflammatory disease,and the effect is correlated with the dose of drug,which may be one of the mechanisms of effect of promoting blood circulation by removing blood stasis on treatment of chronic pelvic inflammatory disease and relief of cervical adhesion by its anti-adhesive effect and inflammation-inhibiting effect.
