摘要
应用响应面法对番茄早疫病拮抗菌WL07的培养基组成及培养条件进行优化。通过Plackett-Burman设计试验,从蔗糖、碳酸钙、硫酸铵、硫酸亚铁、酵母膏、装液量、初始pH等影响因素中筛选影响拮抗菌WL07发酵的显著因素。Plackett-Bur-man设计试验结果表明影响拮抗菌WL07发酵的显著因素分别为初始pH和蔗糖,最陡爬坡试验确定蔗糖浓度3%,初始pH6.5为中心组合设计试验中心点。采用响应面分析法确定了菌株WL07的最佳发酵条件,在蔗糖浓度为30.1 g.L-1,初始pH6.2时,抑菌圈直径增大到原始发酵条件的1.27倍,由原来的22.01 mm增大到28.00 mm,增加约6 mm。
Response surface method was applied to optimize the composition of culture medium and culture conditions of Altermaria solani antagonistic microorganisms WL07.Plackett-Burman experiment was applied to screen the significant factors which could affect on WL07 biological fermentation,from sucrose,calcium carbonate,ammonium sulfate,etc.Plackett-Burman experimental results showed that two significant factors were sucrose and initial pH.The steepest ascent method had determined the concentration of 3% sucrose,the initial pH6.5 as the center combination design experiment center.The optimal fermentation conditions of WL07 could be determined by using the response surface method,when sugar concentration 30.1 g·L-1,initial pH6.2,antibacterial circle diameter was about 1.27-fold increase compared with that using the original fermentation conditions,by 22.01 mm increased to 28.00 mm,increased nearly 6 mm.
