负性协同刺激分子PD-L1在EAE小鼠模型中的表达

摘要 目的:检测PD-L1分子在实验性变态反应性脑脊髓炎(EAE)中的表达,为进一步探讨PD-L1在EAE病理过程中的作用奠定基础。方法:用髓鞘少突胶质细胞糖蛋白(MOG35-55)免疫C57BL/6J小鼠制备EAE模型,进行神经功能评分,对小鼠脊髓进行HE染色、Fast-Blue染色,免疫组化检测脊髓组织PD-L1的表达,并用流式细胞术、Western Blot检测两组动物脾细胞上PD-L1的表达。结果:免疫组化显示实EAE小鼠脊髓PD-L1的表达较对照组明显增加,免疫荧光标记、流式细胞仪计数、Western Blot检测显示实验组动物脾细胞上PD-L1的表达较对照组明显增加。结论:EAE实验组小鼠PD-L1的表达在脾细胞和脊髓都较对照组明显增加。 Objective:To investigate the expression of the costimulatory molecule PD-L1 in the multiple sclerosis model:experimental allergic encephalomyelitis(EAE).Methods:EAE mice were setup by injecting myelin oligodendrocyte glycoprotein(MOG35-55) into the C57BL/6J mice.The clinical manifestation was rated and analyzed.The spinal cord sections from the experiment group and the control group were procedured to H-E staining,Fast-Blue staining.The expression of PD-L1 in the spinal cord was detected by immunohistochemistry.Immuno-flouresence labeling,flow cytometry assay and Western-Blot were carried out to detect the PD-L1 expression on the splenocytes of these mice.Results:The expression of PD-L1 increased in the spinal cord,as well as on the spleen derived CD4+ T cells in the experimental group comparing with that in the control group.Conclusion:The increased expression of PD-L1 both on the CD4+ T cells from spleen and in the spinal cord of EAE group suggested the involvement of the costimulatory molecule in the EAE pathology procedure,which calls on more mechanism explore.
