摘要
目的:了解山茶籽联合雌二醇对去卵巢大鼠骨组织和骨代谢指标的影响,为中西医结合治疗绝经后骨质疏松症提供实验依据。方法:6月龄健康雌性大白鼠120只,分成5个实验组各24只:即假手术组(sham)、去卵巢模型组(OVX)、山茶籽组(Ts)、雌二醇组(E2)、小剂量山茶籽醇提取物+雌二醇组(Ts+E2)。各实验组在第4、8、12和20周随机取6只大鼠取股骨切片观察骨组织,采血测量血钙、磷和碱性磷酸酶,数据进行统计学分析。结果:OVX组的第4、8周血钙、磷明显低于sham组(P<0.01),但OVX组的第12、20周血钙、磷与sham组相比差异无统计学意义(P>0.05)。OVX组的第4、8、12、20周血碱性磷酸酶均明显高于sham组(P<0.01)。3个治疗组各时间的血钙、磷和碱性磷酸酶与sham组差异无统计学意义(P>0.05)。小剂量的雌二醇联合山茶籽能使去卵巢大鼠骨组织和血钙、磷和血碱性磷酸酶基本恢复正常(P>0.05),与较大剂量的山茶籽组或较大剂量的雌二醇组相比无明显差异(P>0.05)。结论:山茶籽醇提取物对去卵巢大鼠的骨质疏松症的治疗效果良好;较小剂量的雌二醇与山茶籽对去卵巢大鼠的骨质疏松症的治疗效果与单独使用较大剂量的山茶籽或较大剂量的雌二醇相比治疗效果相似。
Objective:To understand the effect of camellia seeds combined with estradiol on biochemical indicators of osseous tissue and bone metabolism of ovariectomized rats, provide an experimental basis for treating postmenopausal osteoporosis with combined treatment of Western medicine and Chinese medicine. Methods: A total of 120 six - month healthy female rats were divided into five groups, 24 rats in each group: sham operation group, ovariectomized model group, camellia seeds group, estradiol group, and low dose alcohol extract of camellia seeds plus estradiol group (combined treatment group). Six rats were selected randomly from each experimental group on the fourth, eighth, twelfth, and twentieth weeks to obtain thighbone sections and observe bone tissue, blood samples were obtained to detect the levels of calcium, phosphorus, and alkaline phosphatase, statistical analysis was conducted. Results:The levels of calcium and phosphorus in blood on the fourth and eighth weeks in ovariectomized model group were significantly lower than those in sham operation group ( P 〈 0. 01 ), but there was no significant difference in the levels of calcium and phosphorus in blood on the twelfth and twentieth weeks between ovariectomized model group and sham operation group ( P 〉 0. 05 ). The levels of alkaline phosphatase in blood on the fourth, eighth, twelfth, and twentieth weeks in ovariectomized model group were significantly higher than those in sham operation group ( P 〈 O. 01 ). There was no significant difference in the levels of calcium, phosphorus, and alkaline phosphatase at different time points between the three treatment groups and sham operation group (P 〉 0. 05 ). The osseous tissue and blood calcium, phosphorus, and alkaline phosphatase levels returned to normal in ovariectomized rats treated with low dose estradiol combined with camellia seeds (P 〉 0. 05). Conclusion : The effect of alcohol extract of camellia seeds is good in treatment of ovariectomized rats with osteoporosis; the curative effect of low dose estradiol combined with camellia seeds for ovariectomized rats with osteoporosis is similar to those of large dose camellia seeds alone or large dose estradiol alone.
