快眼动睡眠剥夺、腺苷对大鼠自发活动及海马钙/钙调素依赖蛋白激酶4的影响

摘要 目的:研究快眼动睡眠剥夺(REMSD)、腺苷对大鼠自发活动及海马钙/钙调素依赖蛋白激酶4(CaMKIV)的影响。方法:成年雄性SD大鼠随机分为正常对照组(n=8)和应激造模组(n=48。经21 d慢性轻度不可预见性应激(CUMS)又分为抑郁模型组、REMSD组、水环境对照组、NS对照组、腺苷A1和A2a受体拮抗剂组,每组8只)。用开场实验检测大鼠的自发活动;Western blot检测海马CaMKIV变化。结果:21 d CUMS后,应激造模组总路程、中央路程、周边路程显著减少(P<0.01,P<0.05,P<0.01)。72 h REMSD后,REMSD组与水环境对照组比,总路程、周边路程显著增加(P<0.01);腺苷A1受体拮抗剂组与NS对照组比,总路程、周边路程显著增加(P<0.01)。抑郁模型组CaMKIV较正常对照组显著减少(P<0.01);REMSD组、腺苷A1受体拮抗剂组与抑郁模型组比显著增加(P<0.05,P<0.01);腺苷A2a受体拮抗剂组与抑郁模型组比无统计学意义。结论:REMSD可快速逆转大鼠的抑郁样行为;腺苷、CaMKIV可能参与REMSD抗抑郁机制。 Objective:To study the effects of rapid eye movement sleep deprivation(REMSD)and adenosine on locomotor activity and Ca2+/calmodulin-dependent proteinkinase IV(CaMKIV)level in hippocampus in rats.Methods:Adult male SD rats were divided randomly into normal control group(n=8)and stress model group(n=48).After 21 days chronic unpredictable mild stress(CUMS),the stress model group rats were redivided into the depression-model,REMSD,the tank control,the saline control group,the adenosine A1 and A2a receptor antagonist groups,8 for each group.The locomotor activity in rats was detected by open field test.The changes of CaMKIV level were detected in hippocampus by Western blot.Results:The total distance,middle distance and border distance in the stress model group decreased significantly(P〈0.01,P〈0.05,P〈0.01)after CUMS for 21 days.The total distance and border distance in the REMSD,the adenosine A1 antagonist groups increased obviously after REMSD for 72 hours(P〈0.01).The level of CaMKIV in the depression-model group was significantly lower than that in the normal control group(P〈0.01).The CaMKIV levels in the REMSD and the adenosine A1 receptor antagonist groups were significantly higher than those in the depression-model group(P〈0.05,P〈0.01).The level of CaMKIV had no statistical significance compared with the adenosine A2a receptor antagonist and depression-model groups.Conclusion:REMSD can reverse quickly the depressive-like behavior of rats.Adenosine and CaMKIV may involve in REMSD antidepressant effect.
