寡核苷酸抑制LINGO-1基因表达调控的实验研究

摘要 目的:利用人工合成以LINGO-1基因的mRNA为靶目标的内切磷硫酰核酸序列的寡聚脱氧核糖核苷酸(oligodeoxyribonucleotides,ODN-PSsXT-1),对脊髓损伤部位进行干预,分析LINGO-1表达的改变。方法:采用新生SD大鼠60只制成脊髓损伤模型后,再随机分为ODN-PSsXT-1实验组30只和实验对照组30只进行干预实验;在不同的时间段,利用免疫组化染色,Western blot和RT-PCR方法对细胞表达的LINGO-1 mRNA进行分析。结果:Western blot和RT-PCR分析结果表明,XT-1ODN-PSs可减少LINGO-1基因的表达。结论:人工合成XT-1ODN-PSs寡核苷酸对脊髓损伤后LINGO-1表达有明显的抑制作用,从而有利于脊髓损伤后的再生。 Objective To explore the relationship between oligodeoxyribonucleotide(ODN-PSs) and the injured neurous regeneration by analyzing the expression of LINGO-1 mRNA of astrocyte in vivo interfered by Synthetic phosphorothioate oligodeoxyribonucleotide(ODN-PSs).Methods The spinal cord injuries(SCI) rat models were founded by improved Allen WD,and then randomly divided into experimental group treated with OND-PSs XT-1 and normal control group treated with normal sodium(NS),with 30 rats in each group.The mRNA and protein expressions of LINGO-1 gene and GFAP gene in injured spinal cord were analyzed by Immunocytochemical stain,RT-PCR and western blot techniques.Results The results of western blot and RT-PCR showed that ODN-PSs could down-regulate the mRNA and protein expressions of LINGO-1 gene obviously.Conclusion ODN-PSs can significantly inhibit the expression of LINGO-1,and benefit to the neurous regeneration of injured spinl cord.
