摘要
目的观察8-羟基脱氧鸟苷(8-OHdG)在2型糖尿病模型大鼠肾脏的表达及瑞舒伐他汀钙(Rosuvastatin)对它的影响。方法将大鼠分为正常对照组(C)、糖尿病组(D)、瑞舒伐他汀钙治疗组(N),比较各组大鼠肾组织中丙二醛(MDA)含量、超氧化物岐化酶(SOD)活性,和8羟基脱氧鸟苷(8-OHdG)含量,同时观察血糖、血胰岛素、肾功能和肾脏形态变化。结果与正常组比较,糖尿病组血液中MDA含量明显上升(P<0.05),SOD活性显著下降(P<0.05),8-OHdG增多(P<0.05),肾功能和肾脏形态受损,而瑞舒伐他汀钙治疗组的上述表达较糖尿病组明显改善。结论 8-OHdG在糖尿病肾病中的表达明显增加,而瑞舒伐他汀钙可以通过抑制氧化应激反应对2型糖尿病模型大鼠肾脏产生保护作用。
Objective To evaluate the expression of 8-hydroxyguanine(8-OHdG) in type 2 diabetic rats and the effect of Rosuvastatin on it.Methods 25 rats were divided into 3 groups of normal rats(C),diabetic rats(D),and diabetic rats treated with Rosuvastatin(N).Content of malonaldehyde(MDA) and activity of antioxidant enzymes including superoxid edismutase(SOD) and 8-OHdG in the renal tissue were compared among the groups.In addition,blood glucose,serum creatinine(Cr) and blood ureanitrogen(BUN)were mesured;the renal tissue were observed by light micros copy and electron microscopy.Results Compared with normal group,the level of BUN,Cr,MDA and 8-OHdG in Rosuvastatin untreated group were significantly increased,but the activity of SOD were decreased notably.In addition,the renal pathologic changes in Rosuvastatin untreated group was also decreased,but improved in the treared group obviously.Conclusion Oxidative stress takes key point in the development of diabetic nephropathy.The expression of 8-OHdG in type 2 diabetic rats are increased.Andantioxidation may be the possible mechanism of the nephroprotective action of Rosuvastatin.
