阿托伐他汀对大鼠肥厚心肌组织结缔组织生长因子和核因子-κB的影响 被引量:4

摘要 目的探讨在压力超负荷下阿托伐他汀对心肌肥厚大鼠心肌组织结缔组织生长因子(CTGF)和核因子-κB(NF-κB)表达的影响。方法将40只健康雄性SD大鼠随机分为假手术组(n=10)、模型组(n=10)、阿托伐他汀组(n=10)及卡托普利组(n=10)4组。其中模型组、阿托伐他汀组和卡托普利组采用结扎腹主动脉方法制作心肌肥厚模型。术后24 h阿托伐他汀组给予阿托伐他汀5 mg/(kg.d)、卡托普利组给予卡托普利50mg/(kg.d)溶于5 mL生理盐水灌胃,假手术组和模型组给予等量生理盐水灌胃,6周后检测大鼠部分血流动力学指标、心肌肥厚指数以及心肌组织中CTGF、NF-κB的表达。结果 (1)与假手术组比较,模型组、阿托伐他汀组和卡托普利组舒张末期室间隔厚度(IVST)、左心室舒张末期后壁厚度(LVPWT)、收缩压(SBP)、舒张压(DBP)、左心舒张末压(LVEDP)、心肌肥厚指数和心肌组织CTGF、NF-κB表达明显增高(P<0.05),±dp/dtmax明显降低(P<0.05)。(2)与模型组比较,阿托伐他汀组和卡托普利组IVST、LVPWT、SBP、DBP、LVEDP、心肌肥厚指数和心肌组织CTGF、NF-κB表达明显降低(P<0.05),±dp/dtmax明显增高(P<0.05)。(3)与卡托普利组比较,阿托伐他汀组SBP、DBP、LVEDP明显增高(P<0.05),而IVST、LVPWT、心肌肥厚指数和心肌组织CTGF、NF-κB表达、±dp/dtmax比较差异无统计学意义(P>0.05)。结论阿托伐他汀可延缓心肌肥厚的发展,这一作用可能与其降低心肌组织中高表达的CTGF和NF-κB有关。
